The sale of stocks take about 3 days to "clear" so you would not have access to that cash for about 3 days. If you had other cash in your account, you could of course use that.

But if you can't find a buyer, then no. And it is highly unlikely that the market for your stock will have enough buyers to purchase your 125K shares at $0.10. Penny stocks are notoriously volatile and illiquid.

So if If I bought 125k shares of stock at 0.004 at 9am and then at 10am the stock goes up to 0.1, can I then sell my 125k shares of stock which would be a net of $12.5k

Is this possible?
